I. Immunoglobulin
Immunoglobulin 이란 immune system 에서 antigen (pathogenic bacteria 나 virus)과 특이적 결합을 하여 antigenantibody 반응을 일으키는 물질 이다. 기본 구조는 Y-shaped 의 protein 이며, heavy chain 에 disulfide bond 로 붙어있는 light chain 으로 구성되는 immunoglobin superfamily 에 속 하는 glycoprotein 이다. 5 가지(γ, δ, α, μ, ε) 종류의 heavy chain 이 있으며 이들은 antibody 의 종류를 구분하며, 3 개의 constant region (C_H 1,C_H 2,C_H 3)과 1 개의 variable region(V_H)로 구성된다. Light chain 의 경우 2 가지 종류가 있으며, 1 개의 constant region(C_L)과 1 개의 variable region(V_L)으로 구성된다. IgG, IgD, IgA 는 Hinge 를 통해서 antigen 의 size 에 따라서 조절할 수 있고, IgM, IgE 는 hinge 가 없지 만 additional constant domain 이 있다.
II. B cell maturation
B cell zone (primary follicle)에서 Naïve IgM+, IgD+ B cell 이 antigen 을 만나면 activation 된다. Activated B cell 은 T cell zone 으로 이동하고 TD antigen 이었을 경우, helper T cell 과 bind 되서 T-B interaction 이 일어난다. Helper T cell 의 CD40L 과 B cell 의 CD40 이 bind 되면, helper T cell 에서 cytokine 을 분비하고 이로 인해 B cell 이 proliferation 과 differentiation 을 하여 plasma cell 이 된다.
